The very first DNA ancestry study used only three markers to obtain valid results. As commercial DNA testing for genealogical purposes came on line, the number was gradually increased, so that 37 Y Chromosome Markers are the standard today for male lines. For the same males lines, the YHRD uses 11 markers (9, depending on how you count the "double" markers like DYS389I/II).

For your mitochondrial (mother's female) line, the standard is to test for mutations on both your HVR1 and HVR2 in the so-called D-loop.

For the DNA Fingerprint Test, the standard is the same CODIS markers as seen on TV crime shows.
